English Title: Effect of Endogenous Proteasome Regulation on Cardiac Function and Remodelling in a Murine Hypertrophy Model

Abstract
Das Herzkreislaufsystem ist essentiell für die Aufrechterhaltung lebenswichtiger Organfunktionen wie z.B. die des Gehirns. Die Anpassung der Herzfunktion an eine veränderte transiente oder chronische hämodynamische Belastung initiiert adaptive oder maladaptive Anpassungsreaktionen. Das Ubiquitin-Proteasom-System ist eine zentrale Kontrolleinheit zellulärer Proteine, dessen Manipulation abhängig von ihrem Ausmaß kardiale Umbauprozesse sowohl positiv als auch negativ beeinflussen kann. Die Funktion der fakultativen proteasomalen Untereinheit low molecular mass peptide 2 (Lmp2) bei der Pathogenese kardialer Umbauprozesse ist unzureichend verstanden. Ziel dieser Arbeit war es, den Einfluss von kardialem Lmp2 auf kardiale Umbauprozesse in der Maus umfassend zu bestimmen. Unter basalen Bedingungen entwickelten erwachsene Lmp2 Knock-out-Mäuse keinen offensichtlichen kardialen Phänotyp. Ihre chronotrope, inotrope und lusitrope Reaktion auf kontinuierliche β-adrenerge Stimulation über vier Tage war nicht von der bei Wildtyp-Geschwistertieren zu unterscheiden. Nach anhaltender Stimulation bis insgesamt sieben Tage allerdings waren Verkürzungsfraktion und aktive diastolische Ventrikelfüllung gegenüber der Wildtyp-Gruppe deutlich verringert. Gleichzeitig konnte bei Lmp2 Knock-out-Mäusen eine wesentlich stärkere Zunahme beim Verhältnis von Herz- zu Körpergewicht, bei der Kardiomyozytenquerschnittsfläche, der Hinterwanddicke sowie der interstitiellen Kollagen-Einlagerung beobachtet werden. Die Aktivität des 26S-Proteasoms war bei Lmp2 Knock-out-Tieren weniger stark erhöht. Kardialer Gentransfer von Lmp2 verhinderte den systolischen Funktionsverlust und begrenzte das Ausmaß der kardialen Hypertrophie auf das Niveau der Wildtypmäuse. In einem proteomischen Ansatz mittels zweidimensionaler differenzieller Gelelektrophorese wurden bei Lmp2 Knock-out-Tieren 36 durch Isoprenalin differenziell regulierte Proteine detektiert, von denen 27 im Vergleich zur parallel behandelten Wildtyp-Gruppe gegensätzlich reguliert waren. Zusammengefasst scheint Lmp2 für die myokardiale Entwicklung und basale Herzfunktion nicht notwendig zu sein. Nach hypertropher Stimulation trägt Lmp2 jedoch zur Erhaltung der physiologischen Herzfunktion bei und begrenzt maladaptive Umbauprozesse im Herzen, sodass dieser induzierbaren Proteasomuntereinheit eine kardioprotektive Rolle zugeschrieben werden kann.
Translation of abstract (English)
The cardiovascular system is essential for maintaining proper organ function and viability. Modulation of cardiac function in response to a transient or chronic change in haemodynamic load initiates adaptive or maladaptive cardiac remodelling. The ubiquitin-proteasome system (UPS) is a major regulator of cellular proteostasis. Depending on its extent, manipulation of UPS function can ameliorate or exacerbate cardiac remodelling processes. The impact of the facultative proteasome subunit low molecular mass peptide 2 (Lmp2) on cardiac remodelling is so far poorly understood. Therefore it was the aim of this study to comprehensively determine the impact of cardiac Lmp2 on cardiac remodelling in mice. Unchallenged mice congenitally lacking Lmp2 did not develop an obvious cardiac phenotype. Additionally, their chronotropic, inotropic and lusitropic response to continuous β-adrenergic stimulation for four days was indistinguishable from that of wild type littermates. After a total of seven days stimulation with isoproterenol, though, left-ventricular fractional shortening and active diastolic filling were markedly impaired compared to that found in wild type animals. Decline of cardiac function was accompanied by an augmented gain in heart weight to body weight ratio, cardiomyocyte cross-sectional area and left-ventricular posterior wall thickness as well as by an increased interstitial collagen content and reduced induction of 26S proteasomal activities. Cardiotropic transfer of the Lmp2 gene prevented loss of systolic cardiac function and restricted hypertrophy to level observed in wild type mice. With a proteomic approach utilising two-dimensional differential gel electrophoresis, in Lmp2 knock-out mice 36 proteins differentially regulated by isoproterenol treatment were detected, 27 of which were regulated inversely compared to wild type animals treated in parallel. In conclusion, Lmp2 seems to be non-essential for myocardial development and cardiac function at baseline. In contrast, following hypertrophic stimulation, Lmp2 contributes to maintaining physiological cardiac function and restricts maladaptive remodelling. Hence, a cardioprotective role can be assigned to this inducible subunit of the proteasome.
