English Title: Functional aspects of the two Pex11 homologs Pex11-1 and Pex11-2 concerning peroxisomal biogenesis in mammals

Abstract
Ziel dieser Arbeit war die Funktionsanalyse der Pex11-Homologe Pex11-1 und Pex11-2 aus Säugern, welche integrale peroxisomale Membranproteine mit zwei Transmembrandomänen und in das Zytoplasma ragenden Termini sind. Die funktionelle Bedeutung der Termini, insbesondere des Pex11-2-N-Terminus, sollte geklärt werden. Weiterhin sollte untersucht werden, ob im N-Terminus von Pex11-2 ein Targeting-Signal enthalten ist. Es wurden cDNA-Konstrukte, z.T. mit Verkürzungen am N- und / oder am C-Terminus, und Fusionsproteine mit Marker-Molekülen wie Myc, GFP, CFP und YFP angefertigt. Nach Transfektion der Konstrukte in AT3- und CHO-Zellen wurden diese immuncytochemisch untersucht: I.) wt-Pex11-2-Expression induziert eine etwa Verzehnfachung der Peroxisomenzahl. II.) Myc-Pex11-2-Expression erzeugt in CHO-Zellen tubuläre statt sphärischer Peroxisomen mit intraperoxisomaler Proteinsegregation. III.) GFP-Pex11-2-Expression erzeugt Cluster peroxisomaler Tubuli. Durch Inkubation mit dem Inhibitor Nocodazol wurden Mikrotubuli als maßgebliche Zellstruktur für die peroxisomale Clusterbildung identifiziert. Nach Verkürzung des N-Terminus bleiben bei allen Konstrukten die ursprünglich beobachteten Effekte der unverkürzten Konstrukte aus, obwohl die Proteine in die Peroxisomenmembran inseriert werden. Denkbare Funktionen des Pex11-2-N-Terminus wären: (i) Beeinflussung der Mikrotubuli-Assoziation beim Peroxisomen-Transport, evt. über Kinesin. (ii) Funktion bei der Peroxisomenbiogenese, insbesondere bei der Fragmentierung. Weiterhin zeigen die Ergebnisse, daß die Pex11-Homologe kein funktionsfähiges PTS im N-Terminus enthalten, das wohl eher im luminalen Loop lokalisiert ist. Die Identifikation möglicher cytosolischer Interaktionspartner des Pex11-2-N-Terminus steht noch aus. Die vorliegenden Ergebnisse weisen auf das Vorhandensein unterschiedlicher funktioneller Abschnitte des Pex11-2-Moleküls hin und bilden somit eine Basis für die weitere Charakterisierung dieses Proteins.
Translation of abstract (English)
The aim of this work has been to analyze the function of the two Pex11 homologs Pex11-1 and Pex11-2 in mammals. These are integral peroxisomal membrane proteins with two transmembrane domains and termini which are protruding into the cytoplasm. The functional importance of the termini, especially of the Pex11-2 N-terminus, had to be analyzed. Furthermore there should be elucidated if the N-terminus of Pex11-2 contains a targeting signal. Several cDNA-constructs with deletions of the N-terminus and / or the C-terminus and fusion proteins with Myc, GFP, CFP or YFP were constructed. This was done for truncated and full length constructs. Transfected AT3- and CHO-cells were examined immunocytochemically: I.) wt-Pex11-2-expression induced peroxisomal proliferation by a factor of ten. II.) Myc-Pex11-2-expression in CHO-cells induced tubular peroxisomes instead of spherical ones and also caused intraperoxisomal segregation of proteins. III.) GFP-Pex11-2-expression induced clusters of peroxisomal tubules. Microtubules were identified as the responsible cellular structure for the clustering by incubation with the inhibitor Nocodazol. After expression of N-terminally truncated constructs, there was no change of the peroxisomal phenotype although the protein was inserted into the peroxisomal membrane. Functions of the Pex11-2 N-terminus could be: (i) Influence on microtubule association in the context of peroxisomal transport, e.g. via kinesin. (ii) Importance for peroxisomal biogenesis and the fragmentation of peroxisomes. Further it has been demonstrated that the Pex11 homologs contain no functional PTS in the N-terminus, instead this could be located in the luminal loop. The identification of putative cytosolic interaction partners of the Pex11-2 N-terminus requires further experiments. The results indicate the presence of different functional domains within the Pex11-2 molecule. Hereby they represent the basis for further characterization of this protein.
