Abstract
Zahlreiche epidemiologische, klinische und experimentelle Studien zeigen einen Zusammenhang zwischen der Prostaglandin H-Synthase Cyclooxygenase (COX)-2 und Tumorentwicklung. Mit dem Ziel, die Rolle der COX-2 in der epidermalen Tumorentwicklung zu untersuchen, wurden in dieser Arbeit transgene Mäuse etabliert, in denen eine konstitutive Expression der COX-2-cDNA unter der Kontrolle des Keratin 6 (K6)-Promotors erfolgte (K6-COX-2-Mäuse). Die Stimulierung des K6-Promotors durch 12-O-Tetradecanoyl-phorbol-13-acetat (TPA) bewirkte eine im Vergleich zu Wildtyp-MÄausen länger anhaltende Induktion der COX-2-Expression in der interfollikulären Epidermis transgener Mäuse. In der vorliegenden Arbeit wurden die Auswirkungen dieser transgen-spezifischen Cyclooxygenase-2-Expression auf den Ablauf der Entzündungsreaktion und der Krebsentwicklung gezeigt. Zusätzlich wurden in stratifizierten, Keratin 6-exprimierenden Epithelien morphologische Veränderungen durch eine konstitutive Cyclooxygenase-2-Expression beobachtet
Translation of abstract (English)
A number of epidemiological, clinical, and experimental studies revealed a correlation of the prostaglandin H synthase cyclooxygenase-2 and tumor development. In order to investigate the role of COX-2 in epidermal tumor development, in this study transgenic mouse lines were established with constitutive COX-2 expression under the control of the keratin 6 (K6) promoter (K6-COX-2 mice) in distinct compartments of tongue, esophagus/forestomach, and hair follicles. The stimulation of the K6 promoter by 12-O-tetradecanoylphorbol-13-acetate (TPA) additionally caused a prolonged induction of COX-2 expression in interfollicular epidermis of transgenics as compared to wild-type mice. In this study the effects of transgen-specific cyclooxygenase-2 expression on the inflammatory response and tumordevelopment. In addition, the constitutive expression of cyclooxygenase-2 caused morphological alterations in keratin 6-expressing startified epithelia.
