Abstract

Die Aktivität Natürlicher Killerzellen wird durch eine Vielzahl aktivierender und inhibierender NK Zell Oberflächenrezeptoren reguliert. Die Signaltransduktion der einzelnen Rezeptoren und die Interaktion zwischen den verschiedenen Rezeptoren untereinander sind noch weitgehend unverstanden. Körpereigene Zellen sind durch die Zelloberflächenexpression von MHC Klasse I Molekülen vor einem Angriff der NK Zellen geschützt. Einige Studien lieferten jedoch Daten, die zu der Vermutung führten, dass der aktivierende NKG2D-Rezeptor die Signale der MHC Klasse I-bindenden, inhibierenden Rezeptoren übergehen könnte und trotz der MHC-Klasse I Expression der Zielzellen deren Lyse durch die NK Zellen induziert. In dieser Arbeit wurde zum einen das Wechselspiel zwischen aktivierenden und inhibierenden Rezeptoren am Beispiel des NKG2D-Rezeptors untersucht. So konnte ein Modell über die molekularen Mechanismen der NK Zell Regulation aufgestellt werden. Dabei spielen spezielle Membranbereiche, die sog. Lipid Rafts, eine essentielle Rolle. NKG2D wurde nach Ligandenbindung in diese Membranbereiche rekrutiert. Nach biochemischer Zerstörung der Lipid Rafts war die NKG2D-vermittelte NK Zell Aktivierung blockiert. Es konnte gezeigt werden, dass inhibierende Rezeptoren in die NKG2D Signalkaskade eingreifen, indem sie gezielt die Raft-Rekrutierung des Rezeptors blockieren. Angriffspunkt ist dabei das intrazelluläre Signalprotein Vav1, welches die Reorganisation des Aktin-Zytoskeletts vermittelt. Durch die Stimulation inhibierender Rezeptoren wird Vav1 dephosphoryliert und somit inaktiviert, wodurch die Aktinpolymerisation und die daran gekoppelte Raft-Rekrutierung von NKG2D und die Raft-Akkumulation an der Immunologischen NK Zell Synapse verhindert wird. Die Aktivität der NK Zelle wird nicht nur durch das Zusammenspiel aktivierender und inhibierender Rezeptoren reguliert, sondern auch durch die Modulation der Aktivität von stimulierenden Rezeptoren, wie dem 2B4 Rezeptor. Der 2B4 Rezeptor ist einer der am besten charakterisierten NK Zell Rezeptoren. Er kann NK Zellen direkt aktivieren oder nur als kostimulierendes Molekül fungieren. In unreifen NK Zellen oder in NK Zellen von Patienten mit dem X-linked lymphoproliferative disease (XLP) scheint er sogar inhibierende Signale zu induzieren. Diese Vielseitigkeit der transduzierten Signale könnte durch die vielen verschiedenen intrazellulären Signalmoleküle vermittelt werden, die an den 2B4-Rezeptor binden können. Das kleine SH2 Domänen-haltige Adapterprotein SAP spielt für die Funktion von 2B4 eine essentielle Rolle. XLP Patienten besitzen durch ein mutiertes SAP Gen kein funktionelles SAP Protein und leiden unter einem starken Immundefekt. NK Zellen dieser Patienten können nicht über 2B4 aktiviert werden. Über die Regulation von SAP existieren eine Reihe gegensätzlicher Daten im humanen und murinen System und in unterschiedlichen Zelltypen. In dieser Arbeit wurde deshalb die Regulation von SAP in humanen NK Zellen untersucht und dessen mögliche Auswirkungen auf die Aktivität des 2B4-Rezeptors. Alle Versuche wurden dabei in primären humanen NK Zellen aus gesunden Spendern durchgeführt. Unstimulierte NK Zellen exprimieren kein oder kaum detektierbare Mengen an SAP Protein. Es konnte gezeigt werden, dass die Stimulation humaner NK Zellen mit IL-2, IL-12, IFN-? und polyI:C eine deutliche Hochregulation der SAP-Genexpression induzierte. Interessanterweise konnten unterschiedliche Varianten des SAP Proteins detektiert werden. Desweiteren konnte in dieser Arbeit zum ersten Mal auch eine, im Vergleich zu SAP gegenläufige, Regulation des Adapterproteins EAT-2 gezeigt werden. Die Regulation der SAP-Expression führte zu einer Modulation der 2B4-vermittelten NK Zell Aktivität. So konnte eine klare Korrelation zwischen den exprimierten SAP-Mengen in ruhenden und IL-2 aktivierten NK Zellen und der Aktivität des 2B4 Rezeptors festgestellt werden. Dabei stellte sich heraus, dass 2B4 bei geringen SAP-Mengen eine kostimulatorische Rolle zu übernehmen scheint und in aktivierten NK Zellen die Rolle eines eigenständigen, aktivierenden Rezeptor übernimmt.

Translation of abstract (English)

NK cells are regulated by a variety of activating and inhibitory NK cell receptors. The signal transduction of the individual receptors and the interaction among different receptors are not fully understood. Cells expressing MHC class I molecules are protected against NK cell attacks. In some studies it was suggested that the activating NKG2D receptor can overcome signals from MHC class I binding inhibitory receptors and induces their lysis through NK cells despite the expression of MHC class I. In this thesis the interplay between activating and inhibitory receptors was studied and a model for the molecular mechanism of NK cell regulation was developed. Special membrane regions, the so-called lipid rafts, play an essential role in NK cell regulation. The activating NK cell receptor NKG2D was shown to be recruited into this membrane areas after ligand binding. Biochemical destruction of lipid rafts led to the blocking of NKG2D-mediated NK cell activation. It could be shown that inhibitory receptors could interfere with NKG2D signaling cascade by blocking the raft-recruitment of this receptor. The target of the inhibitory receptors is the intracellular signaling protein vav1 which is important for mediating actin-rearrangement. The stimulation of inhibitory receptors induces a de-phosphorylation of vav1, leading to an inhibition of actin-polymerisation and NKG2D raft-recruitment as well as the blockade of the lipid raft accumulation on the NK cell immunological synapse. NK cells are not only regulated by the interplay between activating and inhibitory receptors but also by the modulation of the activity of stimulating receptors such as 2B4. 2B4 is one of the best characterized activating NK cell receptors. Engagement of 2B4 stimulates NK cells directly but it can also act as a co-stimulatory receptor. In immature NK cells or in NK cells from patients with X-linked lymphoproliferative disease (XLP) 2B4 is also able to transmit inhibitory signals. This variety of transduced signals could be mediated by the binding of different intracellular signaling molecules. The small SH2 domain containing adapter protein SAP plays an essential role for the 2B4 function. XLP patients are characterized by a mutant SAP gene leading to a non-functional SAP protein and resulting in a severe immune defect. NK cells from this patients cannot be activated by 2B4 stimulation. Several conflicting reports in the murine and human system have described a regulated SAP expression in different cell types. Therefore, in this thesis the regulation of SAP expression in human NK cells was examined as well its influence on 2B4 activity. All experiments were done in primary human NK cells. Unstimulated NK cells express little or no SAP protein. Stimulation of human NK cells with IL-2, IL-12, IFN-? and polyI:C induced a strong upregulation of SAP expression. Interestingly, different variants of SAP protein could be detected. Furthermore, a regulation of EAT-2, a SAP-related adapter, could be shown for the first time. The regulated SAP expression induced a modulation of the 2B4-mediated NK cell activity. 2B4 acts as a co-stimulatory receptor in case of low SAP levels whereas it mediates signaling solely in case of high SAP levels. Thus, a clear correlation between the SAP levels in resting and IL-2 activated NK cells and the activity of 2B4 could be detected.
