Abstract

In vitro Pflanzen von H. procumbens, S. nodosa und P. lanceolata wurden aus Samen etabliert und dienten als Ausgangsmaterial zur Etablierung von Kallus- und Wurzelkulturen. Ebenfalls erfolgreich war die Vermehrung der Pflanzen in vitro. Die Iridoidglykoside Harpagosid, Aucubin und Catalpol wurden mittels LC-ESI-MS identifiziert und ihre Anteile in den verschiedenen Pflanzenorganen durch HPLC-Analyse bestimmt. Zum Vergleich wurden intakte Pflanzen analysiert. Die in vitro Pflanzen zeigten unterschiedliche Verteilungsmuster an Iridoidglykosiden. In den in vitro Pflanzen von S. nodosa wurde mit insgesamt 1,68 % (TG) deutlich mehr Harpagosid produziert als in denjenigen von H. procumbens. An Aucubin wurde in den verschiedenen Organen der in vitro Pflanzen von S. nodosa und P. lanceolata ein fast identisches Verteilungsmuster mit einem Gesamtgehalt über 2 % bestimmt. Beim Vergleich mit den intakten Pflanzen wiesen die in vitro kultivierten Pflanzen von S. nodosa nur etwas niedrigere Werte an detektierbaren Iridoiden auf als die intakten Pflanzen (4,36 % bzw. 4,88 %). Die intakten Pflanzen von P. lanceolata zeigten zum Sammelzeitpunkt dagegen einen etwas niedrigeren Iridoidgehalt als die in vitro kultivierten auf (3,70 % bzw. 3,48 %). Die Sekundärstoffproduktion der in vitro Pflanzen konnte durch Zugabe von Methyljasmonat nicht gesteigert werden, aber in den Wurzeln wurde eine Verschiebung des Iridoidglykosidgehaltes zugunsten von Catalpol bewirkt. Beim Vergleich intakter Pflanzen verschiedener Plantago-Arten zeigten sich deutliche Unterschiede bei der Verteilung von Aucubin und Catalpol in den verschiedenen Organen. Kallus- und Wurzelkulturen wurden hinsichtlich ihres Wachstumsverhaltens charakterisiert und auf den Gehalt an Iridoidglykosiden untersucht. Diese waren offensichtlich nicht in der Lage, Iridoidglykoside zu synthetisieren. Hinsichtlich der pharmakologischen Wirkungen wurde für Harpagosid und Harpagophytum-Extrakte in vitro keine Hemmung der COX-1 festgestellt. Eine Inhibierung der LOX aus Sojabohnen wurde für die Iridoidglykoside Harpagosid, Aucubin und Catalpol in schwächerem Maße beobachtet als für verschiedene Pflanzenextrakte von H. procumbens, S. nodosa und P. lanceolata. Im in vivo CAM-Assay am Hühnerembryo konnte für Harpagosid und Catalpol keine antiangiogene Wirkung nachgewiesen werden.

Translation of abstract (English)

In vitro plants of H. procumbens, S. nodosa, and P. lanceolata were established from seeds. Callus and root cultures were established from these plants and plants were regenerated successfully in vitro. Iridoid glycosides, such as harpagoside, aucubin, and catalpol were identified by LC-ESI-MS and their contents determined by HPLC. For comparison intact plants were analysed. The analysis revealed differing patterns of iridoid glycosides in the in vitro plants. In vitro plants of S. nodosa produced remarkably more harpagoside (1.68 %) than those of H. procumbens. For aucubin the pattern of distribution in different organs of in vitro plants of S. nodosa and P. lanceolata was nearly identical with an amount of more than 2 %. In in vitro plants slightly lower amounts of detectable iridoid glycosides (4.36 %) were determined than in intact plants (4.88 %). Intact plants of P. lanceolata contained slightly lower amounts of detectable iridoid glycosides (3.70 %) than in vitro cultivated plants (3.48 %) at time of harvest. Elicitation with methyljasmonate in in vitro plants did not increase production of iridoid glycosides but induced a decrease of aucubin with a simultaneous increase of catalpol in the roots. The comparison of intact plants of different Plantago species revealed remarkable variations in the distribution of iridoid glycosides in the different plant organs. Callus and root cultures were characterised in regard to their growth and analysed for their content of detectable iridoid glycosides. Both were apparently not able to synthesise iridoid glycosides. Concerning the pharmacological effects of harpagoside and Harpagophytum extracts no inhibition of COX-1 could be determined in vitro. An inhibition of LOX from soybeans was achieved by extracts of H. procumbens, S. nodosa, and P. lanceolata in a stronger manner than by the isolated iridoid glycosides harpagoside, aucubin, and catalpol. The in vivo CAM-Assay with chicken embryos revealed no antiangiogenic effects for harpagoside and catalpol.
