Abstract

HIV (human immunodeficieny virus) hat als Erreger von AIDS (acquired immunodeficiency syndrome) 25 Jahre nach seiner Entdeckung bereits zum Tod von weltweit etwa 25 Millionen Menschen geführt. Über zwei Drittel der HIV-Infizierten leben in Afrika. Zwischen weltweit verbreiteten HIV-1-Gruppe M-Viren und nur regional in Afrika vorkommenden HIV-1-Gruppe O-Viren bestehen ausgeprägte genotypische Unterschiede, deren Einflüsse auf Replikation und Pathogenese dieser Primärisolate jedoch weitgehend unbekannt sind. In der vorliegenden Arbeit wurde die humane Tonsillen-Histokultur als physiologisch relevantes experimentelles ex vivo-Modellsystem der HIV-Infektion etabliert. Hierin durchgeführte vergleichende Infektionsstudien zeigten, dass (i) keine grundsätzlichen Unterschiede des Replikationsvermögens zwischen Gruppen M- und O-Viren bestehen, (ii) Gruppe M-Viren sich, in Abhängigkeit von ihrer Korezeptor-Verwendung, untereinander zwar stark in ihrem Replikationsvermögen, nicht jedoch in ihrer Pathogenität (CD4+-T-Zell-Depletion) unterscheiden, und, im Gegensatz dazu, (iii) Gruppe O-Viren sich untereinander stark in ihrer Pathogenität, nicht jedoch in ihrem Replikationsvermögen unterscheiden. Hierbei konnten insbesondere zwei R5-HIV-1-Gruppe O-Viren identifiziert werden, die CD4+-T-Zellen in einem hohen X4-HIV-1-Viren vergleichbaren Maße depletierten. Weiterhin wurde aufgezeigt, dass die bioinformatische Vorhersage der Korezeptor-Verwendung für HIV-1-Gruppe O-Viren mittels geno2pheno[coreceptor] oft fehlerhaft ist, was die Bedeutung phänotypischer Korezeptoranalysen für weniger verbreitete Primärisolate hervorhebt. Trotz immenser Fortschritte bei der Prävention und Therapie von HIV-Infektionen besteht ein dringender Bedarf zur Identifikation und Entwicklung neuer anti-HIV-Wirkstoffe. Im Rahmen dieser Arbeit wurde eine potente anti-HIV-Aktivität in wässrigen Blatt-Extrakten der Lippenblütler Melisse, Pfefferminze und Salbei nachgewiesen und deren Wirkmechanismus analysiert. Es zeigte sich, dass diese Extrakte rasch und nachhaltig die Infektiosität von HIV-1 in nicht-zytotoxischen Konzentrationen verminderten und dadurch die Virus-Fusion beeinträchtigten. Eine Extrakt-induzierte Erhöhung der Viruspartikel-Dichte stellt das wahrscheinliche Korrelat der viruziden Aktivität dar. Weiterführend konnten aus komplexen Lippenblütler-Extrakten einzelne Flavone mit hoher anti-HIV-Aktivität aufgereinigt und näher charakterisiert werden. Diese könnten eine innovative Basis für die Entwicklung neuer topischer Mikrobizide im Kampf gegen HIV darstellen.

Translation of abstract (English)

HIV (human immunodeficiency virus), the causative agent of AIDS (acquired immunodeficiency syndrome), was discovered 25 years ago and has resulted in 25 million deaths worlwide. Two-thirds of HIV-infected individuals are living in Africa. HIV-1 group M viruses have a global distribution, while HIV-1 group O viruses are endemic in distinct regions of Africa. Primary isolates from these HIV-1 groups display pronounced genotypic differences, the consequences of which on the replication capacity and pathogenesis are largely unclear. In this Ph.D. thesis, the human tonsil histoculture was established as a physiologically relevant experimental ex vivo model system of HIV infection. Comparative infection studies conducted herein revealed that (i) no fundamental differences exist between the replicative capacities of HIV-1 group O and M isolates, (ii) group M viruses, depending on the coreceptor usage, widely differ in their replicative ability, but not in their pathogenicity (CD4+ T-cell depletion), and (iii) group O isolates differ markedly in their pathogenicity, but not in their replicative ability. In this context, two R5 HIV-1 group O isolates were identified that caused massive depletion of CD4+ T-cells, to an extent comparable to X4 HIV-1 viruses. Furthermore, the bioinformatic prediction of HIV-1 coreceptor usage of primary O types using the geno2pheno[coreceptor] software proved to be error-prone, emphasizing the importance of phenotypic coreceptor analyses for less prevalent primary isolates. Despite major advances in HIV prevention and pharmacotherapy there remains an urgent need to identify and develop novel anti-HIV agents. A potent anti-HIV activity of aqueous leaf extracts from the Lamiaceae lemon balm, sage, and peppermint was identified and the mode of action was analysed in the framework of this study. These extracts rapidly and effectively reduced the infectivity of HIV-1 virions at non-cytotoxic concentrations resulting in an inhibition of virion fusion. An extract-induced enhancement of the virion density is the likely correlate of the viricidal activity. In addition, individual flavones, purified from complex extracts of Lamiaceae, displayed potent anti-HIV-1 activity. These may provide an innovative basis for the development of topical microbicides in the fight against HIV.
