Abstract

Die pflanzliche Vakuole ist ein multifunktionales Organell, das von zentraler Bedeutung für die Morphologie und Physiologie der einzelnen Zellen und der gesamten Pflanze ist. Aufgrund des hydrolytischen Milieus in ihrem Innern wird sie häufig mit dem Lysosom tierischer Zellen verglichen. Ihre Funktionen sind jedoch wesentlich vielseitiger. Hinzu kommt, dass in den letzten Jahren gezeigt werden konnte, dass eine pflanzliche Zelle zwei funktionell verschiedene Vakuolen aufweisen kann: die lytische Vakuole und die Proteinspeichervakuole. Eine solche Differenzierung, die zwei alternative sekretorische Wege zu den jeweiligen Vakuolen erfordert, wurde bisher weder für das Lysosom tierischer Zellen noch für die Vakuole der Hefe beschrieben. Die zwei verschiedenen Vakuolentypen pflanzlicher Zellen konnten dabei nicht nur aufgrund ihres Inhalts und dessen pH-Wert unterschieden werden, sondern auch anhand verschiedener Isoformen der in ihren Tonoplasten enthaltenen Aquaporine, den „tonoplast intrinsic proteins“ (TIPs). Dabei wird alpha-TIP im Tonoplast als Merkmal für Proteinspeichervakuolen und gamma-TIP als Merkmal für den Tonoplast lytischer Vakuolen angesehen. Ausgehend von dieser Hypothese wurden die Wurzelspitzen von Keimlingen und zehn Tage alten Gersten- und Erbsenpflanzen zunächst ultrastrukturell und dann durch Immunmarkierungen untersucht. Hierfür wurden intakte Wurzelspitzen verwendet, sodass der Entwicklungsstand der Zellen, und damit ebenfalls ihrer Vakuolen, aufgrund ihrer Lage innerhalb der Wurzeln abgelesen werden konnte. Durch die Immuno-fluoreszenz-Markierungen wurde die generelle Verteilung der jeweiligen Proteine innerhalb der Wurzeln ermittelt, während die Immunogold-Markierung bei wesentlich höherer Auflösung der genauen Lokalisation innerhalb der Zellen diente. Diese korrelative Studie ermöglichte somit eine sehr differenzierte Auswertung bei optimaler Auflösung. Dadurch konnte gezeigt werden, dass in Wurzeln von Gerstenkeimlingen und zehn Tage alten Gerstenpflanzen Gerstenlektin gespeichert wird. Bei den Erbsen hingegen wird nur in den Wurzeln von Keimlingen Vicilin gebildet, während in den Wurzeln der zehn Tage alten Erbsenpflanzen Vicilin nicht mehr nachgewiesen werden kann. Dabei konnte eine generelle Korrelation zwischen dem Vorkommen von dem als Markerprotein für den Tonoplast von Proteinspeichervakuolen angesehenen alpha-TIP und den Speicherproteinen in einem Gewebe bestätigt werden. Innerhalb der Wurzeln stellte sich jedoch heraus, dass die Bereiche, in denen alpha-TIP vorkommt, nicht vollständig übereinstimmen mit den Bereichen, in denen die Speicherproteine nachgewiesen wurden. Im Rahmen dieser Arbeit wurde erstmalig eine gleichzeitige Immunogold-Markierung von alpha- und gamma-TIP an Schnitten von Gersten- und Erbsenkeimwurzeln angefertigt. Das Ergebnis dieser Markierung ist, dass in den Wurzelspitzen von Gersten- und Erbsenkeimlingen keine Vakuolen identifiziert werden konnten, die ausschließlich gamma-TIP in ihrem Tonoplast enthalten. Dies steht im Widerspruch zu den Ergebnissen von Immunofluoreszenz-Markierungen von Gersten- und Erbsenkeimwurzel-Quetschpäparaten. Diese führten zu der Theorie, dass in den Gersten- und Erbsenkeimwurzeln zunächst zwei unterschiedliche Vakuolen entstehen, die im Zuge der Entwicklung miteinander verschmelzen. Stattdessen deuten die vorliegenden Daten darauf hin, dass die Vakuolen in den meristematischen Keimwurzelzellen durch autophagische Membranstrukturen entstehen, die zunächst ausschließlich alpha-TIP enthalten. Im Zuge der Entwicklung wird zusätzlich zum alpha-TIP auch gamma-TIP in die Tonoplasten der daraus entstehenden Vakuolen integriert. Da in diesen Vakuolen Speicherproteine nachgewiesen werden können, handelt es sich um Proteinspeichervakuolen. Für das als Markerprotein für lytische Vakuolen angesehene Aquaporin gamma-TIP konnte somit überraschenderweise sehr deutlich gezeigt werden, dass es im Tonoplast von Proteinspeichervakuolen vorkommt. Zusätzlich konnte - entgegen den Erwartungen - in den Tonoplasten der Vakuolen des sich ausbildenden Zentralzylinders, die keine Speicherproteine enthalten und somit lytische Vakuolen darstellen sollten, gamma-TIP nicht nachgewiesen werden. Gamma-TIP kann deshalb in den Wurzelspitzen nicht als Markerprotein für den Tonoplast lytischer Vakuolen eingesetzt werden. Die vorliegenden Daten zeigen somit deutlich, dass zumindest in Wurzelgeweben von der Verwendung der TIP-Isoformen als Merkmal zur Identifikation von verschiedenen Vakuolentypen abgesehen werden sollte und stellt generell in Frage, ob Aquaporine als Marker fungieren können. Um neben den Inhaltsstoffen zuverlässige Merkmale für die einzelnen Vakuolentypen zu finden, wäre es deshalb empfehlenswert in Zukunft nach vakuolären Membranproteinen zu suchen, die an Schlüsselfunktionen der sekretorischen Sortierungsmaschinerie beteiligt sind.

Translation of abstract (English)

The plant vacuole is often considered to be a multifunctional organelle which plays a key role in the morphology and physiology of a single cell and is therefore of great importance for the entire plant. Because of its hydrolytic content it is often compared with the lysosome of animal cells. However, its functions are much more diverse. It has now been shown that a single plant cell can harbour two functionally different vacuoles: the lytic vacuole and the protein storage vacuole. Such a differentiation, which then requires two different secretory pathways, has not been shown either for the lysosome of animal cells or for the yeast vacuole. The different types of vacuoles can be distinguished not only by their contents and pH, but also by different isoforms of tonoplast aquaporins, so called tonoplast intrinsic proteins (TIPs). Currenly, alpha-TIP is regarded as marker for protein storage vacuoles and gamma-TIP as marker for lytic vacuoles. In order to determine how these vacuole types originate and differentiate, the root tips of seedlings and of 10 day old plants of barley and pea were investigated ultrastructurally and by immuno-labelling. Intact root tips were used, so that the developmental stage of the cells and also of their vacuoles could be estimated by their position within the root. By immunofluorescence-labelling the general distribution of the various marker proteins in the roots was ascertained, while the immunogold-labelling was used to investigate the precise localisation within the cells. These correlative studies allowed for a highly differentiated analysis at optimal magnifications. I have been able to show that barley lectin is stored in the roots of barley seedlings and 10 day old plants, whereas in pea the storage globulin vicilin is synthesized only in the root tips of seedlings but could not been detected in the roots of 10 day old plants. In both plants a general correlation between the presence of alpha-TIP and of storage proteins was confirmed. However, there were areas in the root tip where alpha-TIP was present and in which the storage proteins could not be detected. As a part of these studies a simultaneous immunogold-labelling of alpha- and gamma-TIP was archieved for the first time. The most remarkable result of this labelling was that in the root tips of barley and pea seedlings no vacuoles could be identified that contained only gamma-TIP in their tonoplast. This contradicts the results of an immunofluorescence-labelling study made by Paris and co-workers in 1996 on squash preparations of barley and pea seedling root tips. These results led to the hypothesis that in seedling roots of barley and pea two different vacuoles develop side-by-side, which later merge. Instead the data presented in the present work suggest that the vacuoles of meristematic root tip cells develop from autophagic membrane structures that initially only contain alpha-TIP. These membrane structures develop into vacuoles which in addition to alpha-TIP also have gamma-TIP in their tonoplast. Since these vacuoles contain storage proteins, they must nevertheless be classified as protein storage vacuoles. So, surprisingly, the aquaporin gamma-TIP, which hitherto has been regarded as a marker for lytic vacuoles, can also be localized to protein storage vacuoles. In addition, gamma-TIP could not be detected in the tonoplast of vacuoles of the developing central cylinder. These vacuoles do not contain storage proteins and should therefore be regarded as lytic vacuoles. Thus, in those root tips studied here, gamma-TIP is not a marker for the tonoplast of lytic vacuoles. The present data clearly show that, at least in root tips, TIP-isoforms cannot be used as tonoplast markers for different vacuolar types, and that it is questionable if the aquaporins in general can be used as markers for this purpose. To find a true marker for each vacuolar type apart from content markers, it is recommended to look for membrane proteins of the vacuoles that have a key role in the secretory sorting machinery.
