English Title: Analysis of the adjuvant effect of the tegument protein VP22 on the induction of melanoma specific cytotoxic T cells in C57BL/6 mice after DNA immunisation

Abstract
Melanomzellen koennen durch zytotoxische T Zellen, die spezifisch das TRP-2 Differenzierungsantigen erkennen, zerstoert werden. Im Rahmen der vorliegenden Arbeit sollte daher eine TRP-2 DNA-Vakzine hergestellt und die Aktivierung TRP-2-spezifischer T-Zellen in der C57BL/6 Maus nach DNA Immunisierung evaluiert werden. Die Immunogenitaet der Vakzine sollte durch die Fusion von TRP-2 an VP22, dem Tegumentprotein des Herpes Simplex Virus 1, gesteigert werden. VP22 besitzt die Faehigkeit, interzellulaeren Transport von Proteinen zu vermitteln, so dass VP22-TRP-2 Fusionsproteine effizienter in professionelle antigenpraesentierende Zellen (APC), den Initiatoren spezifischer zytotoxischer Immunantworten, transferiert werden sollten. Verschiedene DNA Vakzine wurden hergestellt, die fuer VP22-TRP-2 Fusionsproteine mit jeweils unterschiedlichen TRP-2 Anteilen (gesamtes TRP-2 Protein oder interne TRP-2 Fragmente) kodieren. Durch die Deletion hydrophober N- und C-terminaler TRP-2 Sequenzen sollte ein effizientes Aus- und Einschleusen der Fusionsproteine ermoeglicht werden. Zu Kontrollzwecken wurden entsprechende DNA-Vakzine ohne VP22-Anteil hergestellt. In vivo induzierten VP22-TRP-2 DNA Vakzine tatsaechlich verbesserte TRP-2-spezifische T-Zellantworten: 30 % der VP22-TRP2 immunisierten C57BL/6 Maeuse zeigten eine TRP-2-spezifische T-Zellantwort, waehrend entsprechende Kontrollvakzine (ohne VP22-Anteil) nur in 4 % der Tiere eine TRP-2-spezifische Immunantwort induzierten. Desweiteren konnte mittels Westen Blot Analysen und Immunfluoreszenz-Mikrokopie ein Einfluss von VP22 auf die Stabilitaet beziehungsweise die intrazellulaere Lokalisation fusionierter TRP-2 Proteinfragmente nachgewiesen werden.
Translation of abstract (English)
Melanoma cells can be killed by cytotoxic T lymphocytes specifically recognizing the TRP-2 differentiation antigen. Therefore, a DNA vaccine encoding TRP-2 should be constructed and evaluated for its efficiency to activate TRP-2-specific T cells in C57BL/6 mice. The immunogenicity of the vaccine should be enhanced by fusion of the TRP-2 antigen to the tegument protein VP22 of the herpes simplex virus 1. VP22 can mediate intercellular transport of proteins, thus VP22-TRP-2 fusion proteins should be taken up by professional antigen presenting cells more efficiently. Different DNA vaccines were constructed encoding VP22-TRP-2 fusion proteins with different length fragments of TRP-2 (entire TRP-2 or internal TRP-2 fragments). The deletion of hydrophobic N- and C-terminal TRP-2 sequences should enhance the intercellular transfer of the fusion proteins. As a control, corresponding constructs lacking the VP22 part were generated. In vivo, VP22-TRP-2 DNA vaccines induced TRP-2 specific T cell responses with increased efficiency: 30 % of the mice immunized with different VP22-TRP-2 vaccines exhibited a TRP-2-specific T cell response, while immunization with the corresponding control constructs, lacking the VP22 part, induced TRP-2-specific T cell activation in only 4 % of the mice. Furthermore, Western Blot analysis and immunofluorescence microscopy revealed an influence of VP22 onto the stability and intracellular localization, respectively, of fused TRP-2 proteián fragments.
