Abstract

Diffusionsmessungen mittels Magnetresonanztomographie finden vielfältige Anwendung bei der Untersuchung biologischen Gewebes. Ziel dieser Arbeit war die Entwicklung von Methoden, um aus der nicht-gaußschen Diffusion Rückschlüsse auf die Mikrostruktur einer Probe zu ziehen. Ein indirekt mit den Diffusionshindernissen verbundener Parameter ist die Kurtosis, deren Abhängigkeit von der Diffusionszeit sowie dem Grad der Diffusionseinschränkung theoretisch und experimentell untersucht wurde.

Die neu eingeführte Diffusions-Poren-Bildgebung ermöglicht erstmals eine direkte Bestimmung der Form von geschlossenen Poren oder Zellen unter Verwendung einer Kombination eines langen und eines kurzen Diffusionsgradienten. Das Signal des gesamten Meßvolumens trägt zu einem Porenbild bei; dies löst weitgehend das bei konventioneller MR-Bildgebung auftretende Problem der Signalabnahme bei Verbesserung der Auflösung. Für eine Probe mit verschiedenen Porenformen kann eine gemittelte Porengeometrie gemessen werden. Diese Technik wurde theoretisch hinsichtlich auftretender Bildgebungsartefakte untersucht.

Die Diffusions-Poren-Bildgebung wurde erstmals experimentell demonstriert unter Verwendung von hyperpolarisiertem Xenon-129-Gas, wobei sich eine gute Übereinstimmung mit Simulationen des Diffusionsprozesses zeigte.

Ferner wurde gezeigt, daß eine Diffusions-Poren-Bildgebung auch mit mehreren kurzen Gradientenpulsen realisiert werden kann. Diese Methoden bieten Vorteile hinsichtlich der anwendbaren Sequenztechnik, jedoch Nachteile bezüglich des erforderlichen Signals und des Verhaltens in Gegenwart von Porenverteilungen.

Die Diffusions-Poren-Bildgebung könnte verwendet werden, um nichtinvasiv Parameter wie Zellgröße, Zelldichte oder Axonintegrität zu bestimmen.

Translation of abstract (English)

Magnetic resonance diffusion measurements are widely employed to gain insight into biological tissues. The aim of this work was to develop methods to derive information about the measurement object from the non-gaussian diffusion propagator.

First, the dependence of the diffusional kurtosis, a parameter indirectly related to diffusion restrictions, on the diffusion time and on the degree of diffusion restrictions was investigated.

Second, the diffusion pore imaging technique was introduced, which allows for direct imaging of the shape of closed pores using a combination of a long and a short diffusion gradient. Diffusion pore imaging mainly eliminates the problem of vanishing signal at increasing resolution of conventional MR imaging since an average pore image is detected. Occurring artifacts were investigated and the technique was demonstrated experimentally for the first time using hyperpolarized xenon-129 gas. Simulations and experiments were in good agreement.

Furthermore, methods for an approach to diffusion pore imaging using short gradient pulses only were developed. These methods exhibit advantages regarding the applicable sequence techniques, but disadvantages regarding the required signal and the performance in the presence of pore distributions.

Diffusion pore imaging could be used to noninvasively obtain parameters such as cell size, cell density or axon integrity.
