Abstract

Der Hauptteil der Dissertation beschäftigt sich mit der NMR-Strukturanalyse von drei biologisch relevanten Polypeptiden: Dem Kaliumkanal Ballpeptid Shaker-P22, dem Kaliumkanalporenpeptid von IRK1 und dem Glutathionreduktase - Faltungs-hemmer P11. Es konnte gezeigt werden, dass das Fragments des Shaker K + -Kanals keine wohldefinierte Tertiärstruktur einnimmt. Das 95 Aminosäuren lange Porenpep-tid des IRK1 Kaliumkanals verhinderte aufgrund seiner stark hydrophoben Eigen-schaft eine vollständige, dreidimensionale NMR-Strukturanalyse mit atomarer Auflösung. Jedoch war es durch den Einsatz eines speziellen Präparationsverfahrens erstmals gelungen, signifikante Mengen des Kanalpeptids in SDS-Micellen zu lösen und den Sekundärstrukturgehalt des Peptides mittels NMR-Spektroskopie und CD-Spektroskopie zu bestimmen Die komplette 1 H-NMR-Strukturanalyse des Glutathi-onreduktase - Faltungsinhibitors P11 ergab eine für kurze Peptide seltene, wohldefi-nierte, a-helikale Faltung des Peptids. Weiterhin wird ein neues Verfahren zur Minimierung von t1-Rauschstreifen vorge-stellt. Unter dem Begriff t1-Rauschstreifen sind spektrale Artefakte bekannt, die von diskontinuierlichen Übergängen im Zeitsignal hervorgerufen werden (z.B. Hardware-instabilitäten oder Temperaturschwankungen). Der neue Ansatz zur Reduktion dieser gravierenden Artefakte bestand in der Entwicklung eines Verfahrens in der Zeitdo-mäne, welches mit großem Erfolg an simulierten Spektren getestet wurde.

Translation of abstract (English)

The main part of the thesis is about the NMR structural analysis of three biologically relevant polypeptides: The potassium channel ball peptide Shaker-P22, the potassium channel pore region of IRK1 and the glutathione reductase folding inhibitor P11. It could be shown that the Shaker fragment forms no well-defined structure. The long peptide of the IRK1 potassium channel prevented a complete three-dimensional structural analysis due to its strong hydrophobicity. However it was possible for the first time by the application of a special preparation procedure to solve significant quantities of the channel peptide into SDS micelles and to specify the secondary structure content of the peptide by NMR and CD spectroscopy. The complete 1 H-NMR structural analysis of the glutathione reductase folding inhibitor P11 resulted in a well-defined a-helical folding which is rare for short peptides. A new procedure for the minimization of t1-noise is presented also. The term t1-noise describes spectral artefacts which are caused by intermittent transitions in the time signal (e.g. hardware instabilities or variations in temperature). The new method for the reduction of these serious artefacts resulted in the development of a procedure in the time domain which was tested with success at simulated spectra.
