Abstract
Die 4Pi-konfokale Laserrastermikroskopie besitzt im Vergleich zu herkömmlichen konfokalen Mikroskopen eine 4- bis 7-fach höhere axiale Auflösung. Bisher war sie jedoch auf die problembehaftete 2-Photonen-Anregung (aufwändig, erhöhtes Photobleichen) beschränkt, da nur so eine lückenlose Abdeckung des Raumfre- quenzspektrums durch die optische Transferfunktion (OTF) realisierbar schien. In dieser Arbeit wird die Eignung der 1-Photonen-Anregung in der 4Pi-Mikroskopie theoretisch und experimentell untersucht. Es wird gezeigt, dass im 4Pi-Typ C- Modus, der sowohl die Interferenz des Anregungs- als auch des Detektionslichts von beiden Objektiven nutzt, bei gleichzeitiger Optimierung von Stokes-Shift, Loch- blendengröße und Objektiv-Öffnungswinkel die OTF lückenlos abgedeckt werden kann. Ebenso werden Pupillenfilter untersucht und angegeben, die zur Glättung der OTF führen. Die artefaktfreien Bilder biologischer Strukturen, die mit ei- nem strahlrasternden 4Pi-Typ C-Mikroskop aufgenommen wurden, zeigen eine axiale Auflösung von 70 - 95 nm, was einer Steigerung von 30% gegenüber ty- pischen 2-Photonen-Typ A-Aufnahmen entspricht. Zusätzlich ermöglicht dieses 4Pi-Mikroskop, die axiale Position einzelner Objekte genauer zu bestimmen. Eine Lokalisierungsgenauigkeit von ca. 2 nm wird zur Vermessung der Abstände se- kretorischer Granula zur neuroendokrinen Plasmamembran genutzt und eröffnet neue Ansätze zur Erforschung des Fusionsprozesses intrazellulärer Membranen.
Translation of abstract (English)
Compared to standard confocal microscopes 4Pi-confocal laser scanning micros- copy features a 4- to 7-fold increased axial resolution. However, so far it has been constrained to 2-photon excitation, because this was the only way to obtain a contiguous optical transfer function (OTF). Unfortunately, 2-photon excitation requires complex laser systems and increases photobleaching. In this work the feasibility of 1-photon excitation in 4Pi-microscopy is investigated. By using the interference of the excitation and the detection light of both objective lenses (4Pi- type C mode), as well as by optimising important parameters, such as the Stokes- shift, the size of the detection pinhole and the aperture angle of the objective lens, a continuous OTF has been established also for 1-photon excitation. Dedica- ted pupil filters further smooth the OTF. Images of biological structures recorded with a 4Pi-type C beam-scanning confocal microscope exhibit a resolution of 70 to 95 nm, thus displaying a 30% increase over its 2-photon 4Pi-type A counterpart. The 4Pi-microscope also localises discrete objects more precisely. In a biological application, a localisation accuracy of appr. 2 nm has been used to measure the distan- ces between secretory granules and the neuroendocrine plasma membrane. These studies open up new opportunities for studying membrane fusion in a cell.
