English Title: Development of a technique to determine noninvasively the total content of N-Acetyl-L-Aspartat in the human brain in vivo

Abstract
Lokalisierte 1H-MR-Spektroskopie (MRS) ermöglicht die nicht invasive Messung des Metaboliten N-Acetyl-L-Aspartat (NAA) in vivo im Gehirn des Menschen. Da neuronaler Zellverlust einher geht mit einer Abnahme des NAA-Gehalts, könnte der Verlauf von gehirnschädigenden Erkrankungen mit diesem Parameter verfolgt werden. In dieser Arbeit werden neben einem Vorschlag von O. Gonen et al. eigene Ansätze zur Bestimmung des gesamt-Gehalts an NAA im Gehirn (WBNAA) entwickelt und auf 1,5-T-Ganzkörper-MR-Tomographen implementiert. Die Techniken wurden erfolgreich an Phantomen und Probanden getestet. Im Hinblick auf eine klinische Anwendung wurden zudem verschiedene Methoden der absoluten Quantifizierung entwickelt und experimentell überprüft.
Translation of abstract (English)
Localised 1H MR spectroscopy (MRS) yields a well–resolved signal of N–acetyl–L–aspartate (NAA) in vivo in the human brain. Since NAA is a marker for neuronal cell loss, the progression of degenerative brain diseases may be monitored by determining the whole–brain NAA (WBNAA) content. In this study, MRS techniques that permit selective preparation of the whole-brain in vivo NAA signal were implemented on clinical whole–body tomographs, following an approach due to O. Gonen et al. and own developments. These techniques were successfully tested both on phantoms and volunteers. In addition, in order to obtain a clinically relevant parameter, experiments on different total quantification techniques were performed.
