Abstract
Innovative Methoden der Fluoreszenzmikroskopie liefern die Instrumente für die Analyse der Struktur und Dynamik des menschlichen Genoms auf supramolekularer Ebene. Während dieser Dissertation wurden verschiedene, sich ergänzende Verfahren zur hochpräzisen Größen- und Distanzmessung weiter entwickelt und auf ihre Anwendbarkeit in der Erforschung der Genomnanostruktur untersucht. Die SMI-Mikroskopie macht durch eine räumlich modulierte Beleuchtung Information über Strukturen zugänglich, die wesentlich kleiner als die optische Auflösung sind. Es wird gezeigt, dass ein SMI-Mikroskop auch mit nur einem Objektiv und einem Spiegel realisiert werden kann. Mit diesem Aufbau können an fluoreszierenden Objekten hochpräzise Größenmessungen bis zu einer kleinsten Ausdehnung von 40nm und Distanzmessungen mit einer Präzision von 4nm durchgeführt werden. Als biologische Anwendung wurden erste Schritte zur Analyse der hypoxieinduzierten Regulation des Erythropoetin-Gens unternommen. Zudem wurden die Kalibrationsalorithmen der spektralen Präzisionsdistanzmikroskopie durch die Berücksichtigung lateraler chromatischer Verschiebungen verbessert. Des Weiteren wurde ein Zweiphotonenmikroskop mit schneller Strahlrasterung entwickelt und aufgebaut. Dabei wird ein Weitfeld-Detektor eingesetzt, sodass große Gesichtsfelder bei gleichzeitiger hoher optischer Auflösung aufgenomen werden können. Dieser Aufbau erschließt zusätzlich die spektrale Signatur der Fluoreszenzlebensdauer.
Translation of abstract (English)
Innovative methods of fluorescence microscopy provide the tools for the analysis of the structure and dynamics of the human genome on the supramolecular level. In this thesis various complementary approaches to measure sizes and positions very accurately were improved and their applicability to biological studies was addressed. SMI microscopy uses a spatially modulated illumination to access information about structures well below the resolution limit. It is shown that SMI microscopy can also be implemented with only one objective lens and a mirror. With this setup high precision size measurements of fluorescent objects down to a smallest extension of 40nm and distance measurements with a precision of 4nm are feasible. In a biological application first steps towards an analysis of the hypoxia induced regulation of the erythropoietin gene expression were taken. Furthermore, the calibration algorithms of spectral precision distance microscopy were enhanced by considering lateral chromatic shifts. Additionally, a two-photon microscope with very fast beam scanning was developed and assembled. A widefield detection scheme is used to simultaneously record a large field-of-view at very high resolution. This setup gives also access to the spectral signature of the fluorescence lifetime.
