Abstract

Kernpunkt dieser Arbeit ist die Entwicklung eines Sensorsystems zur medizinischen Diagnostik von Immunreaktionen. Ein großes Interesse besteht an einer schnellen Detektion von Antigen-Antikörper-Reaktionen. Geringe Stoffkonzentrationen sollen im Bereich von µg/ml in Plasma, Serum oder Blut nachgewiesen werden. Im Rahmen dieser Arbeit wurde ein einfach handhabbares Sensorsystem entwickelt, das für den Einsatz in Arztpraxen und Laboratorien geeignet ist. Als physikalische Grundlage dienen Sensoren auf der Basis von Oberflächenwellen. Als Meßgröße wird die Schallgeschwindigkeit detektiert, die sich aufgrund der Anlagerung des nachzuweisenden Stoffes an der sensitiven Bauteilschicht ändert. Ein Schwerpunkt dieser Arbeit ist eine Steigerung der Sensitivität des Sensors in Hinblick auf die bereits am Markt etablierten Systeme. Zur Führung der Oberflächenwelle wird hier erstmals Spin-On-Glas verwendet, welches wesentliche Vorteile gegenüber den sonst üblichen Materialien bietet. Besondere Interesse besteht darin, eine chemische Ankopplungsmethode zu entwickeln, die eine unspezifische Anlagerung von unerwünschten Stoffen aus Serum oder Blut verhindert. Dies gelang mit dem hier erstmals in der Sensorik verwendeten Polymer Polyvinylamin, das aus seinem Einsatz in der Papierindustrie bekannt ist.

Translation of abstract (English)

This work is focused on the development of a sensor system for the medical diagnostics of immunological reactions, where a high interest exists in the rapid detection of antigen-antibody reactions. Low concentrations in the range of µg/ml should be detectable in plasma, serum, or blood. In this work a sensor system has been developed that is simple to operate and suited for the use in medical practices or laboratories. The sensors are based on surface acoustic waves, the measured quantity being the sound velocity which is influenced by an adsorption of the analyte at the sensitive layer of the device. A central point of this work is the enhancement of the sensitivity of the sensor relative to that of systems already established on the market. For the first time a spin-on-glass has been employed to guide the surface wave, offering significant advantages over the commonly used materials. Special interest exists in the development of a chemical coupling technique that prevents the unspecific attachment of other components of the serum or blood sample. This could be achieved using the polymer polyvinyl amine which is known from its use in the paper industry and which has been used here for the first time in a sensor application.
